Description:
The multidrug resistance to a variety of
structurally unrelated cancer chemotherapeutic
agents often results in the
failure of cancer treatments. The multidrug
resistance-associated protein (MRP) gene
family contains at least 6 members
designated MRP1 to 6. MRP1 is a
phosphoprotein of 1531 amino acids and is
expressed in a variety of cell types. MRP1
mRNA has been demonstrated in lung,
testis and peripheral blood mononuclear
cells but was not detected in placenta, brain,
salivary gland, liver, uterus and spleen. The
protein has been detected immunohistochemically
in the epithelium and glands of
nasal respiratory mucosa as well as in 44
percent of cases of ovarian cancer. The
clinical relevance of MRP-mediated multidrug-
associated resistance in human
cancers is still not understood. Increased
levels of MRP mRNA have been noted at
high frequency in blood samples of patients
with chronic lymphocytic leukemia but no
relation to prior chemotherapy or treatment
was observed. Clone 33A6 will be of
interest in studying the expression of
multidrug resistance-associated protein 1 in
a broad range of tumors.
Specificity:
Recognizes human multidrug resistanceassociated
protein 1.
Species Reactivity:
Human. Others not tested.
Ig Isotype:
Mouse IgG2a
Immunogen: Recombinant prokaryotic protein
corresponding to a cytoplasmic domain,
between transmembrane regions 5 and 6, of the
human MRP1 molecule.
Positive control:
Bowel.
Format:
Lyophilized tissue culture supernatant containing
15 mM sodium azide. Reconstitute in 1 mL of
sterile distilled water.
Storage and Stability:
Store unopened lyophilized antibody at 4°C.
The reconstituted antibody is stable for at least
two months at 4°C. For long-term storage, store
aliquots of the antibody at -20oC to avoid freezethaw
cycles. Prepare working dilutions on the
day of use.
Applications and Suggested Dilutions:
 Immunohistochemistry: (paraffin sections)
Use at a 1:25–1:50 dilution.
High-temperature antigen unmasking
technique. Incubate with the primary antibody
for 60 minutes at 25oC. Standard ABC
technique.
Staining Pattern: Cytoplasmic.
The optimal dilution for a specific application
should be determined by the researcher.
Limitations:
For in vitro research use only. Not for use in
diagnostics or in humans.
